芸香对人膀胱癌EJ细胞体外作用的研究

摘要 目的:探讨芸香对人膀胱癌EJ细胞的作用。方法:MTT法检测EJ细胞生长抑制率;流式细胞仪检测细胞周期改变和免疫组化SP法检测Ki-67蛋白表达的变化。结果:芸香浓度为30、60、90μmol/ml时,EJ细胞的生长受到不同程度的抑制,且具有时间和浓度依赖性。EJ细胞经不同浓度的芸香作用24h后G0/G1期细胞比率明显下降,而G2/M期比率明显上升。EJ细胞Ki-67的阳性率随芸香浓度的增高而下降(P<0.05)。结论:芸香能抑制癌细胞的生长增殖。其机制可能通过诱导癌细胞周期阻滞于G2/M期、下调Ki-67表达有关。 To study antitumor mechanisms of quercetin on human bladder cancer cell line EJ in vitro. Methods:Cell suppression rates were treated by MTT assay, cell cycle was measured by flow cytometry and Ki-67 expression was detected by immunohistochemistry. Results:Different levels of cell suppression rates were observed while EJ cell were treated with different concentrations(30.60.90/mol/ml) of quercetin, and in a time and concentration dependent manner. The ratio of Go/Gl phase cells was decreased and the ratio of G2/M phase cells was increased, when treated with 30.60.90 μmol/ml quercetin for 24 h. The expression of Ki-67 was down-regulated when treated with 30.601.90 μmol/ml quercetin. Conclusions: Quercetin could inhibit human bladder cancer EJ cell growth. The possible mechanism was to induce cell cycle arrest at G2/M phase and down-regulate the expression of Ki-67 protein.
