肾癌及癌旁组织蛋白质组学的比较研究 被引量:1

摘要 目的:建立肾癌及癌旁组织比较蛋白质组学双向电泳技术研究体系。方法:取病理证实的肾癌及癌旁组织,研磨、超声裂解、抽提蛋白,采用双向凝胶电泳分离,进行染色,Image Master 2D Platinum软件分析、识别差异表达蛋白质,应用基质辅助激光解吸电离飞行时间质谱得到相应肽质量指纹图谱,搜索数据库鉴定部分差异蛋白质点。结果:从双向电泳图谱上获得蛋白质斑点:肾癌组织(1326±78)个,癌旁组织(1232±56)个。显著差异蛋白质点46个,对其中7个点进行了筛选鉴定。结论:分析这些差异蛋白有助于寻找具有普遍性意义的肾癌标志物,进而为肾癌的早期诊断和防治提供理论和实验依据。 Objective: To establish two-dimensional eleetrophoresis profiles with high resolution and reprodueibility from renal cell carcinoma (RCC) tissues and paired tumor-adjacent kidney tissues and to identify the differential expression proteins. Method: The total proteins of RCC tissues determined by histological and paired tumor-adjacent kidney tissues were separated by two-dimensional gel electrophoresis(2-DE), analyzed by Image Master 2D software, and identified by peptide mass fingerprint(PMF) and database searching. Result: Well-resolved, reproducible 2-D maps of the above tissues were obtained. Some of the different proteins identified by mass spectrometry(MS) were matched in the SWISS-2DPAGE database. Condusion: These data will be valuable for studying the diagnostic markers, mechanisms of metastasize and therapeutic targets.
