电针内关对心肌缺血再灌注损伤大鼠心肌细胞内钙离子浓度的调节 被引量:21

摘要 目的:与电针神门和合谷进行比较,以观察缺血再灌注损伤大鼠电针内关后心肌细胞内钙离子浓度的变化。方法:实验于2003-11/2004-02在湖南中医药大学针灸推拿系完成。选用雄性Wistar大鼠50只,体质量200~250g,购自河南医科大学实验动物中心,随机分为假手术组、模型组、内关组、神门组及合谷组,每组10只。采用经典的冠脉结扎法建立大鼠心肌缺血再灌注模型,双波长荧光分光光度计检测心肌细胞内钙离子浓度。结果:大鼠50只全部进入结果分析。大鼠缺血再灌注后心肌细胞内钙离子浓度增加,以模型组最高,明显高于假手术组(P<0.01)。经电针治疗各组细胞内钙离子浓度降低,神门组和内关组均明显低于模型组(P<0.05,0.01);而合谷组与模型组比较,差异不明显(P>0.05)。内关组细胞内钙离子浓度明显低于神门组和合谷组(P<0.01)。结论:①抑制心肌细胞内钙超载可能是电针内关对缺血再灌注损伤心肌保护的作用机制之一。②内关与神门疗效的差异性提示手厥阴心包经和心之间有相对特异性联系。 AIM: To study Ca^2+ concentration in cardiomyocytes in rats with ischemia/reperfusion injury after electro-acupuncture at Neiguan (PC 6) and compare with that after electro-acupuncture at Shenmen (HT 7) and Hegu (LI 4). METHODS: The experiment was performed at the Department of Acupuncture and Moxibustion, Human University of Traditional Chinese Medicine from November 2003 to February 2004. Totally 50 male Wistar rats with the body mass of 200-250 g were purchased from Laboratory Animal Center of Henan Medical University and were randomly divided into sham operation group, model group, Neiguan group, Shenmen group and Hegu group with 10 in each group. The animal model of myocardium ischemia/reperfusion was set up by ligation of coronary artery. Ca^2+ concentration in cardiomyocytes was detected by dual wave spectrofluorometer. RESULTS: Totally 50 rats were involved in the result analysis. Ca^2+cencentraUon in cardiomyocytes was elevated after ischemia/reperfusion, and was the highest in the model group, which was obviously higher than the sham operation group (P 〈 0.01 ). Ca^2+ cencentraUon in cardiomyocytes was degraded in each group after electro-acupuncture. It was significantly lower in the Shenmen group and Neiguan group than the model group (P 〈 0.05, 0.01). No significant difference was found between the Hegu group and the model group (P 〉 0.05). Ca^2+ concentration in cardiomyocytes was markedly lower in the Neiguan group than the Shenmen group and the Hegu group (P 〈 0.01). CONCLUSION: ①Electro-acupuncture at Neiguan may protect myocardial tissue after ischemia/reperfusion injury through inhibiting calcium overload in the cardiomyocyte. ②The difference between Neiguan and Shenmen curative effect indicated that relative specific association between pericardium channel of hand jueyin and cardiomyocyte was found.
