摘要
试验共接种1851个幼穗,对36份材料离体培养反应进行研究,结果表明,不同基因型在幼穗出愈速度和出愈率、分化速度和分化率方面均存在明显差异。但出愈和分化之间并禾发现相关关系。且出愈及分化速度与材料的熟期早晚无关。试验还表明,花粉母细胞减数分裂期并非幼穗培养的临界期。
The study was carried out in 1984~1988 in Harbin.36 cultivars,lines and F from distant hybridization were nsed in the study. 1851 young inflorescences from plants at booting stage were cultured. Plant regenera-tion from Cultured inflorescences occurred throngh or-ganogeneses, The media used in the study were MS and N_6.The results of study showed that there was significant difference between genoiypes in relation to the frequency and speed of callus formation and plant regeneration. Genotyp played an important role in the success or failure in tissue culture,The study indicated that the ability of callus intdnction, root and shoot differentiation, and plan regeneration depending on genotype was not only found in embryo culture bnta lso in inflorscence culture.The results of study also indicated that no relationships between frequency and speed in callus formation and plant regenerntion were found,The maturity of genotype did not influence the speed of callus induction and plant regeneration.The young inflorescences at the stage as late as after meiosis could be induced callus formation and root and shoot differentiation, which is quite different from the conclusion drawn by other researchers. Some genotypes at this developmental stage had very ihgh frequency of callus induction and root and shoot differentiation in the study.
