摘要 目的:探讨表皮生长因子(epiderm algrowth factor,EGF)与胰岛素对小鼠体外受精(in vitro fertilization,IVF)胚胎植入前发育的影响。方法:小鼠IVF胚胎培养在添加不同浓度的EGF和/或胰岛素-葡萄糖的CZB培养基中,于hCG注射后138h检测囊胚发育率及每个囊胚的细胞数目。结果:培养基中添加0.1ng/mlEGF显著提高了小鼠扩张囊胚的形成率,而囊胚的细胞数目并未增加。EGF与胰岛素-葡萄糖共同添加对小鼠IVF囊胚发育率及囊胚细胞数目并未有显著提高。结论:EGF通过刺激细胞分化,而非细胞增殖方式促进小鼠IVF胚胎的早期发育。而各自最佳浓度的EGF与胰岛素共同添加对小鼠早胚并未有协同促进作用。 Objective: To evaluate influence of epidermal growth factor(EGF ) and/or insulin on the preimplantation development in vitro of mouse in vitro fertilization (IVF} embryo. Methods: Mouse IVF embryos were cultured in the CZB media supplemented with EC, F at different concentrations and/ or insulin - glucose. Blastocyst formation rate and the cell number per blastocyst were examed 138h after human chorionic gonadotropin (hCG ) injection. Results: Supplementation of media with EGF at the concentration of 0. lng/ml caused a significant increasing in the rate of expanded blastocyst formation, but no difference in cell number per blastocyst. Addition of EGF and insulin - glucose in the media did not promote the preimplanta- tion development of mouse IVF embryos. Conclusion : EGF can promote the development in vitro of mouse preimplantation embryo derived from IVF by stimulating the cell differentiation, but not proliferation. However, insulin and EGF at respectively optimal concentration have no synergistic effect on the preimplantation development of mouse IVF embryos.
