摘要
目的建立用毛细管电泳对3,5-双三氟甲基苯乙醇对映体的手性拆分方法。方法采用环糊精及其衍生物作为手性选择剂,用毛细管电泳技术对3,5-双三氟甲基苯乙醇对映体进行分离,考察了环糊精种类、环糊精浓度、缓冲溶液pH值、缓冲溶液浓度、分离电压、温度等因素对分离的影响,并对分离条件进行了优化和重复性考察。结果在pH=8.0的磷酸缓冲溶液(0.05 mol.L-1)中,以0.01 mol.L-1的磺丁基-β-环糊精为手性选择剂,15 kV为分离电压,在15℃下可使3,5-双三氟甲基苯乙醇对映体达到基线分离,分离度为1.54。分离度RSD小于2%,迁移时间RSD小于5%。结论该方法简单快速,灵敏度高,重复性好。
OBJECTIVE To establish the method for the separation of 3,5-Bis(trifluoromethyl) phenyl ethanol by capillary electrophoresis. METHODS Using β-cyclodextrin and its derivatives as chiral selectors. The effects of the type and the concentration of cyclodextrin, buffer pH, buffer concentration, voltage, and temperature used on resolutin were studied and optimized. The reproducibility were also investigated. RESULTS Using 10 mmol· L^-1 sulfobutyl ether-β-cyclodextrin as the chiral selector, and 0.05 mol · L^-1 phosphate solution( pH 8.0) as the running buffer,the base line separation of 3,5-Bis(trifluoromethyl) phenyl ethanol enantiomers was achieved with a resolution factor of 1.54. The RSD for resolution and migration time were lower than 2% and 5%. CONCLUSION The developed method is sensitive and accurate ,and can be repeated easily.
