摘要 目的研究Echistatin对人增生性瘢痕成纤维细胞增殖、凋亡及Ⅰ、Ⅲ型前胶原mRNA表达的影响。方法体外培养人增生性瘢痕成纤维细胞,设Echistatin处理组和对照组,MTT法、流式技术及RT-PCR法检测人增生性瘢痕成纤维细胞增殖、凋亡及Ⅰ、Ⅲ型前胶原mRNA的表达。结果Echistatin可明显抑制人增生性瘢痕成纤维细胞增殖,促进凋亡,降低Ⅰ、Ⅲ型前胶原mRNA的表达。结论Echistatin可作为防治增生性瘢痕的筛选药物。 [Objective] To investigate the effect of echistatin on proliferation, apoptosis, and type Ⅰ and type Ⅲprocollagen mRNA expression in human hypertrophic scar-derived fibroblasts. [Methods] After treated by eehistatin, proliferation, apoptosis, type Ⅰ/type Ⅲ procollagen mRNA expression in cultured fibroblasts from human hypertrophic sear were determined by MTT method, Flow eytometry , and RT-PCR respectively. [Results] Eehistatin treatment resulted in significant inhibition of proliferation, type Ⅰ/type Ⅲ proeollagen mRNA expression, and marked apoptosis in cultured hypertrophic scar-derived fibroblasts. [Conclusion] Eehistatin may be chosen as a drug to prevent and cure hypertrophic scar.
