化痰活血软坚法干预肾小球硬化机理的实验研究 被引量:2

摘要 目的:探讨化痰活血软坚法防治肾小球硬化的作用机理。方法:将Wistar大鼠随机分为正常组,模型组和中药组。除正常组外,其余大鼠制成肾小球硬化大鼠模型。中药组大鼠灌胃中药(1mL/100g),其余两组灌胃等量蒸馏水,共12w。然后观察各组大鼠肾组织TGF-β1mRNA、TGF-β1及MMP-9mRNA的表达。结果:与模型组比较,中药组大鼠肾组织TGF-β1和TGF-β1mRNA表达显著降低(P<0.01),MMP-9mRNA表达显著升高(P<0.01)。结论:减少肾组织TGF-β1的产生和分泌、促进MMP-9mRNA表达是化痰活血软坚法发挥效应的重要机理,而抑制TGF-β1mRNA的表达是减少TGF-β1产生和分泌的原因之一。 Objective: To explore the mechanism of therapy of dissipating phlegm, promoting blood flow and removing stasis on treating glomerular sclerosis. Methods:Wistar rats were randomized into normal group, model group and herb group. Rat models of glomerular sclerosis were established. Then herb group rats were given herb( 1 mL/100 g)by stomach perfusing, normal group and model group were given equal dose distilled water for 12 consecutive weeks. The expressions of TGF-β1, TGF-β1mRNA and MMP-9mRNA in rat nephfidial tissue were detected by method of immunoiluorescence and hybridization in situ. Result:Compared with model group, the expressions of TGF-β1 and TGF-β1 mRNA in nephridial tissue of herb group rats lowered obviously(P〈0.01 ), the expression of MMP-9mRNA ascended obviously(P〈0.01 ). Conclusions: The inhibition of TGF-β1 expression and promotion of the MMP-9mRNA expression maybe the important mechanism of the therapy of dissipating phlegm, promoting blood flow and removing stasis on treating glomerular sclerosis. The repression of TGF-β1mRNA expression causes the decrease of TGF-β1 expression.
