化痰活血软坚法干预肾小球硬化机理的实验研究

Mechanism Study of Therapy of Dissipating Phlegm,Promoting Blood Flow and Removing Stasis on Treating Glomerular Sclerosis

目的:探讨化痰活血软坚法防治肾小球硬化的作用机理。方法:将Wistar大鼠随机分为正常组,模型组和中药组。除正常组外,其余大鼠制成肾小球硬化大鼠模型。中药组大鼠灌胃中药(1mL/100g),其余两组灌胃等量蒸馏水,共12w。然后观察各组大鼠肾组织TGF-β1mRNA、TGF-β1及MMP-9mRNA的表达。结果:与模型组比较,中药组大鼠肾组织TGF-β1和TGF-β1mRNA表达显著降低(P<0.01),MMP-9mRNA表达显著升高(P<0.01)。结论:减少肾组织TGF-β1的产生和分泌、促进MMP-9mRNA表达是化痰活血软坚法发挥效应的重要机理,而抑制TGF-β1mRNA的表达是减少TGF-β1产生和分泌的原因之一。

Objective： To explore the mechanism of therapy of dissipating phlegm, promoting blood flow and removing stasis on treating glomerular sclerosis. Methods：Wistar rats were randomized into normal group, model group and herb group. Rat models of glomerular sclerosis were established. Then herb group rats were given herb（ 1 mL/100 g）by stomach perfusing, normal group and model group were given equal dose distilled water for 12 consecutive weeks. The expressions of TGF-β1, TGF-β1mRNA and MMP-9mRNA in rat nephfidial tissue were detected by method of immunoiluorescence and hybridization in situ. Result：Compared with model group, the expressions of TGF-β1 and TGF-β1 mRNA in nephridial tissue of herb group rats lowered obviously（P〈0.01 ）, the expression of MMP-9mRNA ascended obviously（P〈0.01 ）. Conclusions： The inhibition of TGF-β1 expression and promotion of the MMP-9mRNA expression maybe the important mechanism of the therapy of dissipating phlegm, promoting blood flow and removing stasis on treating glomerular sclerosis. The repression of TGF-β1mRNA expression causes the decrease of TGF-β1 expression.
