摘要
为得到产高酶活木聚糖酶的生产菌株,以黑曲霉A3为出发菌,经紫外线亚硝酸复合诱变处理,使所产木聚糖酶酶活由原来的9.1829 U/ml增加到15.2144 U/ml。实验通过发酵培养时间,碳源含量,接菌量对液体发酵产木聚糖酶的影响分析,得出产木聚糖酶的最优条件,即在加入50 Mandels液体培养基,28℃条件下,底物含量为0.045g,接种量为20ml,培养36h。
In order to get a strain with high xylanase activity, Aspergillus niger A3 was treated by UV-ray and nitrous acid. The xylanase activity was increased to 15.2144 U/ml from 9.1829 U/ml. Fermentation time, carbon content and inoculum influence to the liquid state fermentation were studied. Results showed optimal conditions to produce xylanase as follows: the liquid containing 70 ml, temperature 28℃, the bottom thing measures 0.045g of deal, inoculate deal a density 5×106s/ml spore liquid 20ml, educate time 36 hours.
