摘要
目的用固相方法合成血管紧张素Ⅱ,建立其纯化和分析条件。方法以Fmoc-Phe-Wang树脂为固相载体,采取Fmoc/tBu正交保护固相合成策略,粗品的纯化采用制备型RP-HPLC。结果经过分析型RP-HPLC分析最后纯品的纯度>98%,并用电喷雾接口质谱分析证实了分子质量的正确性。结论用Fmoc固相法合成血管紧张素Ⅱ快速简便、操作方便而且回收率很高,最后通过RP-HPLC纯化,可以得到纯度很高的血管紧张素Ⅱ。
Objective To synthesize angiotensin Ⅱ by solid phase method, and to establish the conditions of purification and analysis. Methods Angiotensin Ⅱ was synthesized by Fmoc/tBu orthogonal protection strategy on Fmoc-Phe-Wang resin. The crude peptide was purified using preparative RP-HPLC. Results The purity of the final product confirmed by analytical RP- HPLC was more than 98%. Electrospray interface mass spectrometric analysis showed the right molecular mass of the synthetic peptid.Conclusion The Fmoc solid phase method is simple and easy in the synthesis of angiotensin Ⅱand the yield of the final product is high. The pure angiotensin Ⅱthrough the preparation of RP-HPLC has been obtained at last.
