高效液相色谱法测定穿黄消炎片中穿心莲内酯和脱水穿心莲内酯的含量

Determination of andrographolide and dehydroandrographolide in Chuanhuang Xiaoyan tablets by HPLC

目的 建立HPLC测定穿黄消炎片中穿心莲内酯和脱水穿心莲内酯含量的方法。方法 色谱柱为Dikma Diamonsil C18柱（4．0mm×250mm，5μm），以甲醇-水（50：50）为流动相，脱水穿心莲内酯检测波长254nm、穿心莲内酯检测波长为225nm；采用外标法定量。结果 穿心莲内酯与脱水穿心莲内酯的线性范围内分别为2．928～146．4μg·mL^-1和5．98-299μg·mL^-1，相关系数r均为0．9999；回收率分别为101．82％（RSD=2．2％）和97．52％（RSD=2．4％）。结论 本方法操作简便、重现性好，适合于本制剂的含量测定。

Objective To establish a method for the determination of andrographolide and dehydroandrographolide in Chuanhuang Xiaoyan tablets by HPLC. Methods Chromatography was performed on a Dikma Diamonsil C18 column （4. 6 mm ×250 mm, 5μm）. The mobile phase was methanol - water （50 ： 50）, the detective wavelength of andrographolide and dehydroandrographolide was UV 225 nm and 254 nm respectively. Results The linear range of the determination of andrographolide was 2. 928-146.4μg . mL^-1 , while that for dehydroandrographolide was 5.98-299μg . mL^-1. The average recovery of andrographolide was 101.82% , while that for dehydroandrographolide was 97. 52%. RSD of andrographolide was 2.2%, and that for dehydroandrographolide was 2.4% （n=5）. Conclusion The method is simple, reliable, and suitable for the quality control of the drug.
