结直肠癌MMP与P53表达研究及意义

Study of the expression and significance of MMP and P53 in colorectal adenocarcinoma

目的:探讨MMP7、MMP13和P53在结直肠腺癌中表达及意义。方法:用免疫组化PV9000法检测61例结直肠腺癌和61例癌旁结直肠粘膜组织MMP7、MMP13和P53表达情况。结果:结直肠腺癌组织中MMP7、MMP13和P53表达强度显著强于癌旁结直肠粘膜组织(P<0.01);MMP7在结直肠腺癌中的表达强度与组织学分级、浸润深度、临床病理分期、淋巴道转移有关(P<0.05),与发病部位及远处转移无关(P>0.05);MMP13的表达强度与TNM分期、淋巴道转移及术后5年内死亡有关(P<0.05),与发病部位、组织学分级、浸润深度、远处转移及术后复发转移无关(P>0.05);P53表达强度与术后复发转移有关(P>0.05)。结论:MMP7、MMP13和P53在结直肠腺癌的发生及淋巴道转移中可能起重要作用,测定MMP7、MMP13和P53有助于判断其预后,MMP7、MMP13和P53强阳性者预后差。

Objective To study of the expression and significance of MMP7, MMP13 and P53 in colorectal adenocarcinoma. Methods The expression of MMP7, MMP13 and P53 was examined by PV9000 immunohistochemical technique in sixty- one cases of colorectal adenocarcinomas and 61 cases of cotorectal tissues near to carcinomas. Results The expression of MMP7, MMP13 and P53 were stronger in which colorectal adenocarcinomas than in which with normal colorectal tissues . Of the colorectal adenocarcinomas , the positive intensities of MMP7 was associated with the histological grade and clinicopathological stage as well as lymph node metastasis diseases. Of the colorectal adenocarcinomas, the expression of MMP13 was associated with the lymph node metastasis diseases and poor survival （ P 〈0.05）, but not correlated with the tumour position, histological grade, tumour depth and distant metastasis disease （ P 〉0.05）, those with lymph node metastasis disease and poor survival had increasingly MMP13 expression than those without lymph node metastasis and good survival. P53 expreeion was crrelated with distant metastasis and recur . Conclusions MMP7, MMP13 and P53 maybe play an important role on the development and lymph node metastasis of colorectal adenocarcinomas. It is an useful marker for predicting the prognosis of colorectal adenocarcinomas.
