益母草对糖尿病肾病大鼠血液流变学及转化生长因子-β_1表达的影响 被引量:20

Effects of Leonurs Heterophyllus Sweet on blood rheology and transforming growth factor-β_1 in rats with diabetic nephropathy

摘要 目的:观察益母草对糖尿病肾病大鼠血液流变学及转化生长因子-β1的影响.方法:取健康SD大鼠48只.随机均匀分为正常组、模型组、益母草组、黄芪组4组.益母草组、黄芪组分别灌服5g/(kg·d)益母草、黄芪水煎液,正常对照组、模型对照组灌服相同体积生理盐水.造模前禁食12h.将链脲佐菌素(STZ)用0.1mmol/L,pH为4.4的枸椽酸缓冲液配成1%的浓度,以50mg/kg的剂量一次性腹腔注射,同时喂养高脂高糖半合成高热量饲料,2wk后测定空腹血糖,血糖值超过16.65mmol/L者,确定为模型成功.所有大鼠均自由饮食,分别于造模和杀检时测量体质量,并于杀检时称量肾质量,用代谢笼收集24h尿液.实验8wk后,大鼠在戊巴比妥钠麻醉下,腹主动脉取血并灌洗肾脏,将肾脏分为两部分,一部分于-80℃冰箱保存,另一部分用中性福尔马林固定.运用免疫组化法及血液流变仪分别观察益母草对糖尿病肾病大鼠转化生长因子-β1的表达及血液流变学的影响.结果:益母草水煎液对糖尿病肾病大鼠肾组织中TGF-β1蛋白表达有下调趋势,但无统计学意义(P>0.05),而对其血流变学指标具有明显改善作用.结论:益母草对糖尿病肾病防治作用可能与其改善血流变学指标有关. AIM: To observe the effects of Leonurs Heterophyllus Sweet (LHS) on blood rheology and transforming growth factor-β1(TGF-β1) in rats with diabetic nephropathy. METHODS: A total of 48 SD rats were randomly divided into 4 group: normal control group, model group, LHS treated group and Radix Astragali treated group. The latter 2 groups were given 5 g/( kg·d) LHS or Radix Astragali extract, and the first 2 groups were administered same-volume normal saline. Diabetic nephropathy models were induced by injecting streptozotocin (STZ) in 0. 1 mmol/L buffer (pH 4.4), and fed high-lipid, high-glucose diets. Two weeks later, the animals with fasting blood glucose level of over 16. 65 mmol/L were selected as models. Their body mass was weighed at modeling and sacrifice, and the kidneys were taken for weighing, divided into two parts, half for refrigeration at - 80℃ and half for fixing in formalin. 24 h urine was collected as well. lmmunohistochemlstry was applied to detect the expression of TGF-β1 in nephridial tissue and blood rheometer was employed to determine the change of blood rheology. CONCLUSION: Leonurs Heterophyllus Sweet had a protective effect on diabetic nephropathy, which may be related to its improving the index of blood rheology.
