耐热糖化酶的分离纯化及部分性质 被引量:3

摘要 采用微波诱变筛选出产生耐热糖化酶的突变株,其粗酶液的最适作用温度比出发株高15℃;采用SDS-Starch-PAGE测定出突变株含有3个耐热糖化酶组分;利用DEAE-Cellulose-52柱层析和制备电泳纯化了3个耐热糖化酶组分,各组分的最适温度分别为,60℃,65℃和65℃,分子量分别为136ku、56.3ku和49.3ku。 A mutant which produces thermostable glucoamylase was obtained through microwave mutation, the optimum temperature of the mutant crude enzyme was 15 ℃ higher than that of primitive strain; A convenient method of SDS-Starch-PAGE was established by which three thermostable glucoamylase components were identified, these components were purified through DEAE-Cellulose-52 chromatography and preparative electrophoresis, the optimum temperature of these components were 60 ℃, 65 ℃,65 ℃,their Molecular Weight were also determined by SDS-PAGE and they were 136 ku,56.3 ku and 49.3 ku respectively.
