摘要
目的:研究与丝/苏氨酸蛋白激酶Plk1(polo-like kinase1)相互作用的分子。方法:通过酵母双杂交技术初步确定可能与Plk1存在相互作用的靶分子,进一步通过蛋白分子的细胞亚定位、体内免疫共沉淀和GST-pull-down分析Plk1与候选蛋白之间的相互作用。结果:细胞亚定位表明,Plk1与转录阻遏子NAC1(transcriptional re-pressor nucleus accumbens-1,transcriptional repressor NAC1)在空间上存在相互作用的可能,酵母双杂交、体内免疫共沉淀、GST-pulldown分析均表明Plk1和NAC1存在相互作用。结论:Plk1和NAC1存在相互作用,二者的相互作用可能在细胞的发育分化、肿瘤及神经系统疾病的发生发展中起着重要作用。
Objective: To investigate the protein partners interacted with the polo-like kinase 1 (Plk1). Methods: The mouse embryonic stem cells cDNA library was screened by yeast two-hybrid assay, one of the candidate molecules was found to be nucleus accumbems-1 ( NAC1 ), The subcellular location of NAC1 and Plk1 was determined by fluorescent microscopy, The interaction of Plk1 and NAC1 was confirmed by co-immunoprecipitation in vivo and GST-pulldown in vitro, Results and Conclusion:The data indicated that Plk1 could interact with NAC1 both in vitro and in vivo. The interaction between Plk1 and NAC1 may play an important role in the cellular development ,particularly in the onset and development of the oncological and nervous system diseases.
