摘要
目的观察体外三磷酸腺苷(ATP)对骨髓问充质干细胞(MSCs)生物学特性的影响。方法抽取兔骨髓,采用淋巴细胞分离液密度梯度法分离单核细胞,体外培养传代,观察不同代细胞的生长特性;在第三代时加入ATP.观察MSCs的形态学变化,并行诱导细胞的组织化学染色。结果MSCs在体外可以扩增,以第三代细胞的增殖最快,在ATP存在的条件下向神经样细胞分化,巢蛋白、神经元特异性烯醇化酶及胶质纤维酸性蛋白抗体免疫组化染色呈阳性。结论MSCs在体外ATP的作用下分化为神经细胞。
Objective To investigate the effects of adenosine triphosphate (ATP) on the biological features of bone-marrow mesenchymal stem cells (MSCs) of rabbit in vitro, Methods Bone marrow was aspirated and MSCs were isolated with lymphocyte separating liquid by density gradient centrifuge and cultured in vitro. The 3rd passage of MSCs was incubated with ATP and the morphological changes during incubation period were observed. The induced differentiation of MSCs was identified by immunohistochemical staining. Results Bone marrow MSCs differentiated into nerve cells and expressed nestin, neuro-specific enolase and glial fibriliary acidic protein with the induction of ATP in vitro. Conclusion When incubated with ATP, MSCs can differentiate into nerve cells in vitro.
