摘要
木糖发酵是木质纤维素发酵的关键,休哈塔假丝酵母是自然界木糖发酵性能较好的天然酵母之一。研究了装液量、转速、木糖浓度、有机氮源等条件对休哈塔假丝酵母CICC 1766发酵木糖生产乙醇的影响,探讨休哈塔假丝酵母的乙醇耐受能力。结果表明,休哈塔假丝酵母对酒精的耐受能力达3%vol。在28℃,初始pH值5.0,160r/min,250mL三角瓶装液量150mL,木糖浓度60g/L,玉米浆0.5%(v/v),含尿素0.024%的条件下,休哈塔假丝酵母发酵木糖能产乙醇20.52g/L,得率达到理论值的74.5%。
Xylose fermentation is the key factor for lignocellulose fermentation. Candida shehatae is one of the best xylose-fermenting yeast in nature. In this paper, the influence of liquid loadage, rotating speed, xylose concentration and organic nitrogen source on xylose fermentation by Candida shehatae CICC 1766 to produce ethanol were investigated. Besides, ethanol tolerance of Candida shehatae CICC 1766 was studied. The results showed that ethanol tolerance of Candida shehatae was 3 %(v/v). Ethanol yield could reach 20.52 g/L (74.5 %of theoretical yield) under the following conditions: fermentation temperature at 28 ℃, initial pH value as 5.0, 150 mL liquid loadage in 250 mL rotating flask, and rotating speed at 160 r/min, and 0.5 % (v/v) corn steep liquor and 0.024 % urea.
