摘要
目的研究与蛋白激酶CK2B发生相互作用的蛋白。方法应用酵母双杂交系统,以人蛋白激酶CK2B为诱饵,筛选成人肝cDNA文库,利用生物信息学分析,一对一酵母回复性杂交等提供的信息,筛除假阳性。结果经过酵母双杂交共获得211个克隆,经过验证分析,最终确定12个相互作用蛋白,其中3个是已知的,9个是新发现的相互作用。结论获得的9个新的相互作用蛋白可能揭示CK2新的作用机制。
Objective To study the interaction proteins of CK2B by yeast two-hybrid system. Methods Using the CK2B as bait, a human liver cDNA library was screened and the proteins interacting with CK2B were searched. The false clones were eliminated by bioinformatic methods and by one to one yeast two-hybrid system. Results Twelve interactions were obtained after verification, concluding three known interactions and nine new. Conclusion The encoding proteins obtained may play important roles in the new function of CK2B.
