Establishment and evaluation of rat models of type 2 diabetes

摘要 目的建立类似人类2型糖尿病大鼠模型。方法♂大鼠90只,随机取20只为对照组,70只为高脂组。在脂肪乳灌胃5个月后行高胰岛素-正常血糖钳夹实验,测体重、收缩期血压、空腹血糖、空腹胰岛素、甘油三酯。高脂组(62只)大鼠腹腔注射小剂量链脲佐菌素和弗氏完全佐剂每周一次至造模成功。结果①5个月后,高脂组体重、收缩压、血清甘油三酯较对照组显著升高(P<0.05),空腹胰岛素明显升高,胰岛素敏感指数明显下降,差异有显著性(P<0.05)。②高脂组葡萄糖灌流速度较对照组下降,差异有显著性(P<0.05)。结论长期高脂饮食结合腹腔注射小剂量链脲佐菌素和弗氏完全佐剂成功复制出实验性2型糖尿病大鼠模型。 Objective To establish a rat model similar to human type 2 diabetes mellitus (T2DM) . Methods 90 male SD rats were randomly divided into control group (n = 20) and high-fat diet (HF)group (n = 70), were fed with high-fat emulsion via gavage for 5 months, insulin sensitivity were evaluated by using euglycemie hyperinsulinemie clamps technique and body weight(BW) ,systolic blood pressure (SBP) ,fasting blood sugar (FBG) ,fasting insulin (Fins) serum triglyeride (TG) were measured. Small dose of streptozotoein (STZ) and complete Freund's adjuvant (CFA) were given to the remain HF rats via intraperitoneal injecting once a week until they were diagnosised type 2 diabetes. Results ① After 5 months, BW,SBP,serum TG in HF group were significantly higher than those in the control group( P 〈0. 05 ) ,Fins was signifieantly higher than that in the control group,insulin sensitivity index was significantly lower than that in the control group (P 〈 0. 05 ). ② The HF group acquired significant lower GIR compared with the control group ( P 〈 0. 05 ). Conclusion Intraperitoneal injecting low dose of STZ and CFA after long time of high-fat diet to rats may induce the type 2 diabetes that is similar with human.
