摘要
[目的]研究中药慢肾1号方对IgA肾病小鼠肾小球系膜区血小板源性生长因子(PDGF)及转化生长因子β1(TGFβ1)表达的影响。[方法]采用口服小牛血清清蛋白(BSA)及酸化水感染的复合法建立小鼠实验性IgA肾病模型,用免疫组化方法检测慢肾1号方对该模型肾小球系膜区内PDGF、TGFβ1因子的表达。[结果]本实验在IgA肾病小鼠肾组织系膜区内检测到PDGF、TGFβ1表达,且与肾脏系膜区的病理损伤呈正相关,提示PDGF、TGFβ1参与并介导了IgA的发生、发展。肾高治疗组PDGF、TGFβ1的表达及病理损伤弱于模型组,组间差异具有显著性意义(P<0.01,P<0.05);其中模型组系膜区PDGF、TGFβ1的表达及病理损伤高于正常对照组,与各组间差异具有显著性意义(P<0.01);正常组中PDGF、TGFβ1的表达较弱,与其他各组的差异均有显著性意义(P<0.01);肾高治疗组PDGF、TGFβ1的表达及病理损伤弱于肾低治疗组且具有显著性意义(P<0.05)。[结论]慢肾1号方对IgA肾病小鼠肾小球系膜区PDGF、TGFβ1表达有抑制作用,而抑制系膜区PDGF、TGFβ1的表达可能是慢肾1号方治疗IgA肾病的有效机制之一。
[ Objective] The research of the MS1 decoction which influence the IgA nephrosis mouse PDGF and TGFβ1 expresses. [ Methods] Using the compound methods of oral administration calf blood serum albumen (BSA) and the acidified water infection established the mouse IgA nephrosis experimental model , using the immunochemist method to examine the model renal glomerulus which is in the membrane area PDGF, TGFβ1 the factor expression due to the MS1 decoction. [ Result] In this experiment ,We can find the expresses of the PDGF and the TGFβ1 in the membrane area of the IgA nephrosis mouse kidney organization, and it is related with the damage of the pathology damage in the kidney membrane area. It Prompted that the PDGF, TGFβ1 participate the occurrence and the development of the IgA . In Kidney higher treatment group, the PDGF, TGFβ1 expression and the pathology damage is weakly than in the model . two groups difference has the significance statistics significance ( P 〈 0. 01, P 〈 0.05 ) ; In the model group, the PDGF, TGF β1 expression and pathological injury in the mesangial area is stronger than the normal control group, and the difference between the groups is significant statistical significance;In the normal group, the expression of PDGF , TGF - β1 is weaker and the other group differences are statistically significant ; In Kidney higher treatment group, the PDGF, TGFβ1 expression and the pathology damage is weakly than in the Kidney lower treatment group . two groups difference has the significance statistics significance (P 〈 0.05 ). [ Conclusion] To the expression of the IgA nephrosis mouse' PDGF, TGFβ1, The MS1 decoction has the inhibitory action . It is one of the possible treatment of IgA nephrosis effective mechanisms by using the MS1 decoction that it can be inhibit the expression of the membrane area PDGF, TGFβ1.
