摘要
目的:研究环孢霉素A(CsA)对体外培养的球后成纤维细胞增殖的影响。方法:用MTT法测定CsA对体外培养的正常人和甲状腺相关眼病(TAO)患者球后成纤维细胞增殖的影响。结果:经1,10,100,500,1000μg/LCsA作用48h后,正常人球后成纤维细胞的A值分别为0.253±0.018,0.210±0.016,0.173±0.032,0.134±0.013,0.119±0.022与对照组0.278±0.019相比,差异有统计学意义(P<0.05);经1,10,100,500,1000μg/LCsA作用48h后,TAO患者球后成纤维细胞的A值分别为0.221±0.025,0.191±0.036,0.150±0.017,0.118±0.039,0.110±0.011与对照组0.249±0.028相比,差异有统计学意义(P<0.05);随着浓度的增加,抑制作用增强。结论:CsA呈浓度依赖性抑制正常人和TAO患者球后成纤维细胞增殖。
AIM: To investigate the proliferative effects of cyclosporin A (CsA) on cultured human retroocular fibroblast growth.
METHODS: Effects of CsA on cultured human retroocular flbroblast growth were measured with mythyl thiazolyl tetrazolium (MTT) assay in normal people and patients with thyroid associated ophthalmopathy (TAO).
RESULTS: The A values of 1, 10, 100, 500 and 1 000μg/L CsA were 0.253±0.018, 0.210±0.016, 0.173±0.032, 0.134± 0.013 and 0.119±0.022 after 48 hours in fibroblasts of normal subjects, with significant difference compared with that of the control (0.278±0.019, P〈0.05). The A values of 1, 10, 100, 500 and 1 000μg/L CsA were 0.221±0.025, 0.191± 0.036, 0.150±0.017, 0.118±0.039 and 0.110±0.011 after 48 hours in retroocular fibroblast of TAO patients, with significant difference compared with that of the control (0.249± 0.028, P〈0.05). The inhibitive effect of CsA grew with the increase of its concentration.
CONCLUSION: The CsA has dose-dependent inhibitive effects on the fibroblests proliferation of both normal people and TAO patients.
