益气化瘀方对大鼠腰神经根损伤后生长相关蛋白-43与蛋白基因产物9.5的作用 被引量:3

Effect of the Yiqi Huayu Recipe on Growth-Associated Protein-43 and Protein Gene Product 9.5 Following Lumbar Nerve Root Compression in Rats

摘要 目的:探讨益气化瘀方在神经再生修复过程中对神经肌肉接头部(NMJs)生长相关蛋白-43(growth-associatedprotein-43,GAP-43)及蛋白基因产物9.5(protein gene product 9.5,PGP9.5)的作用。方法:大鼠L5神经根损伤后,给予益气化瘀方分别治疗10 d3、0 d和60 d。采用免疫组织化学和激光共聚焦扫描显微镜技术,观察GAP-43与PGP9.5在大鼠比目鱼肌中的分布变化。采用α-金环蛇毒萤光结合剂显示乙酰胆碱受体(acetylchline receptor,AchR),NIH图像分析系统定量测定GAP-43与PGP9.5重叠面积。结果:益气化瘀方组GAP-43和PGP9.5在NMJs的表达及两者的重叠面积均明显优于模型组。结论:GAP-43与PGP9.5的表达与AchR的神经支配状况密切相关。益气化瘀方能明显加快神经再生修复进程。 Objective:To study the effect of Yiqi Huayu Recipe on growth-associated protein-43 (GAP-43) and Protein Gene Product 9.5(PGP 9. 5)in neuromuscular junctions during regeneration. Methods: Rats with lumbar nerve root compression were given Yiqi Huayu Recipe for 10 , 30 and 60 days respectively. The distribution of GAP-43 and PGP 9.5 in neuromuscular junctions of soleus muscles were observed by immunohistoehemieal method and eonfoeal laser scanning mi- croscopy technique. The acetylchline receptor (AChR) was visualized by fluorescein-conjugatedα-bungarotoxin (α- BTX). The overlap areas of GAP-43 and PGP9.5 was measured by NIH image technique. Result:The expression of GAP- 43 and PGP9.5 in the neuromuscular junctions and the overlap areas of GAP-43 and PGP9.5 in the Yiqi Huayu Recipe group were both better improved than those in the untreated group. Conclusion:The expression of GAP-43 and PGP9.5 are related to the state of innervation for muscles. Yiqi Huayu Recipe can accelerate this nerve regeneration process.
