摘要
采用活体大鼠肝门静脉灌流法,初步探讨了硫酸镍肝损伤的机制,从氧化应激的角度探讨了硫酸镍一次性门静脉染毒后30、60、120、180min时大鼠肝损伤。结果:血清中谷丙转氨酶(GPT)的活性显著增高,镜下可见肝细胞充血、水肿、坏死。不同时间血中丙二醛(MDA)的含量明显增高,同时伴有还原型谷胱甘肽(GSH)的耗竭,并呈剂量-时间反应关系。提示镍导致肝脏氧化应激反应,造成急性肝损害。
In this paper,the mechanism of hepatic injury induced by nickel sulfate in rats was investigated through vivi perfusion of hepatic portal.Meanwhile,the induced hepatic injuries of rats at 30min,60min,120min and 180min after toxicant exposure were discussed,respectively.Besides,the remarkable increases in the activity of serum GPT and the content of serum MDA were observed,and a decrease in the content of GSH was also discovered,which show a dose time response relationship.These experimental results indicated that nickel sulfate may miduce an oxidation stress response and even a acute hepatic injury in rats.
