病理性瘢痕中结缔组织生长因子的免疫组化研究 被引量:20

摘要 目的:研究结缔组织生长因子在病理性瘢痕组织中的表达,以探讨其与病理性瘢痕形成的关系。方法:分别留取正常皮肤15例,浅表性瘢痕20例,增生性瘢痕、增生性瘢痕边缘正常皮肤、瘢痕疙瘩及瘢痕疙瘩边缘正常皮肤各20例组织标本,应用HE染色、免疫组化(SABC法)染色,观察CTGF在各组织中的表达,并用图像分析系统定量分析其在不同组织中表达水平的差异性。结果:CTGF在增生性瘢痕、瘢痕疙瘩、瘢痕疙瘩边缘正常皮肤中的表达均明显高于正常皮肤,有显著性差异(P<0.05);而在浅表性瘢痕、增生性瘢痕边缘正常皮肤中仅有极微弱的CTGF阳性着色,与正常皮肤相比差异无统计学意义(P>0.05)。免疫组化研究显示CTGF主要定位于真皮层成纤维细胞胞浆中。结论:结缔组织生长因子与病理性瘢痕临床生物学行为之间有相关性,在其发病过程中可能发挥重要作用,可能成为今后临床治疗瘢痕的一个有力靶位。 Objective To explore the relation between CTGF and pathologic scar by studying the expression of CTGF in pathologic scar. Methods The expression intensity of CTGF were examined with HE staining ,immunohistochemistry (SABC method) for parts in 15 normal skins,20 shallow scars,20 hypertrophic scars lesions and the adjacent uninvolved skins ,20 keloid lesions and the adjacent uninvolved skins . Then the distribution was analysised quantitatively using image analytical system in various tissue. Results Compared with normal controls,the mean levels of CTGF expression were significantly increased in hypertrophic scars, keloid lesions and adjacent uninvolved skin (P〈0.05),and were very slender in shallow scars, the adjacent uninvolved skins of hypertrophic scars,but there is no statistically significant between them(P〉 0.05).The immunohistochemistry study confirms that the expression of CTGF is mainly in fibroblast cytolymph of dermis. Conclusion CTGF has dependability in the clinic biological behavior with pathologic scar and play an important functions on the formation of pathologic scar and may be a new potential target for the therapeutic intervention of scar later.
