摘要
目的:研究小鼠腹腔多次注射不同剂量STZ对KM种小鼠血液学及胰岛细胞的影响.方法:SPF级昆明种雄性小白鼠(24~28 g),随机分为阴性对照组和3个STZ腹腔注射组,分别为45、35、25 mg/kg·bw剂量组(n=15).各剂量组腹腔多次注射STZ,每次注射后进行空腹血糖检测,并于实验结束时进行血ALT、AST、TP、ALB含量、尿糖含量及PLT、MPV、PCT、PDW测定,同时取胰腺进行组织病理学检查.结果:45 mg/kg·bw剂量组腹腔注射STZ一次,35、25 mg/kg·bw剂量组注射两次,均可显著升高实验动物血糖值(P<0.05);45、35 mg/kg·bw剂量组注射三次后,诱导糖尿病动物模型的成模率分别为93.3%、73.3%,两组的AST、ALT、PLT、尿糖值均高于阴性对照组,但TP、ALB含量低于阴性对照组(P<0.05);胰腺组织病理学检查表明STZ可损伤胰岛细胞,并以45 mg/kg·bw剂量组胰岛细胞损伤最为严重.结论:腹腔注射STZ三次可损伤胰岛细胞,35 mg/kg·bw以上剂量可明显改变实验动物的血生化指标和PLT水平,45 mg/kg·bw剂量的STZ可诱导较为理想的糖尿病动物模型.
Objective :To explore the effects of STZ on blood indice and islet cells changes of mice after being multiply injected intraperitoneally. Methods: The 24 - 28 g KM male mice of SPF were randomly divided into control group and three STZ - treated groups (45,35,25 mg/kg·bw separately) by their weight( n = 15). Animals in STZ groups were multiply injected with STZ intraperitonially, and Glu was measured after each injection. When finishing the experiment, blood indice including ALT, AST,TP, ALB, urine glucose, and PLT,MPV,PCT,PDW were assayed, and pathological changes of pancreas were examined through HE staining. Results:STZ was injected intraperitoneally for once in 45 mg/kg · bw group, twice in 35 mg/kg·bw group, could significantly increase Glu level(P 〈0. 05). After injection for three times,STZ dose of 45 mg/kg· bw and 35 mg/kg·bw could successfully induce diabetes model (93.3% and 73.3% ), and increase the level of ALT, AST, PLT and urine glucose, but lower the level of TP and ALB(P 〈0. 05). Pathological observation of pancreas showed islet cells injury in 45 mg/kg ·bw group were injured the most. Conclusion:Multiple injected STZ intraperitonially for there times could injure islet cells, and STZ above 35 mg/ kg·bw promotes indice of blood biochemistry and PLT changed obviously. The 45 mg/kg · bw of STZ injected three times intraperitoneally could make an ideal diabetes model.
