反相高效液相色谱法检测保健食品中洛伐它汀 被引量:1

摘要 目的:建立可行、准确的液相色谱方法分析检测不同类型保健食品中的功效成份洛伐它汀.方法:在酸性介质中用三氯甲烷提取试样,通入氮气在50℃水浴下挥去三氯甲烷,用流动相定容至一定体积,过膜,滤液10 μl进机作HPLC分析测定.色谱条件:Inertsil 4.6×250 mm C18反相柱,紫外检测波长238 nm,流动相甲醇:水=4:1用磷酸调节pH=3,流速1.0 ml/min,柱温:室温.结果:本法回收率为90.5%~93.8%,RSD=1.36%,洛伐它汀标准溶液浓度在2.0~300.0 μg/ml范围内与峰面积呈良好的线性关系,相关系数为0.9995,检出限:0.2 mg/100 g.结论:本法准确可行,适用于保健食品中的功效成份洛伐它汀的检测. Objective:To establish a method for determination of lovastatin in health foods by reverse phase high performance liquid chromatography with ultraviolet detection. Methods:The lovastatin samples were added into H3 PO4 (pH = 3 ), then extracted with methanol or trichloromethane and detected by RP - HPLC. The chromatographic conditions included a ODS - Cis 250 mm × 4. 6 mm column and using methyl alcohol : H2O = 4 : 1 ( pH = 3 ) as mobile phase. The flow rate was 1.0 ml/min. The decetive wavelength was set at 238 nm. Results:The limit of this method was 0. 2 mg/100 g. The recovery was 90. 5% -93.8% and the RSD was 1.36%. Conclusion:The method is sensitive, accurate and it can be used as a quantitative analytical method for lovastatin in health foods.
