摘要
目的:研究沙眼衣原体(CT)对女性生殖道感染患者局部生殖道上皮免疫状态的影响。方法:(1)选取2006年4月至8月被诊断为生殖道沙眼衣原体感染患者及无生殖道感染的生育年龄女性,每组各60例,取其阴道灌洗液,用ELISA方法比较IL-8,TNF-α,HD-5水平;(2)采用与沙眼衣原体类似来源的脂多糖(LPS),刺激体外培养的正常宫颈上皮细胞,检测LPS刺激后不同时间点TNF-α,HD-5水平。结果:沙眼衣原体组与正常组相比,阴道灌洗液IL-8,TNF-α,HD-5均升高,有明显差异。体外实验结果表明,LPS刺激体外培养的宫颈上皮细胞,0、24、48h细胞培养液中TNF-α,HD-5水平均增加,且呈现时间依赖性。结论:沙眼衣原体感染可影响女性生殖道局部免疫状态,体外实验证实宫颈上皮细胞可能参与了上述过程。
Objective: To investigate the effect of chlamydia trachomatis (CT) on the immunological responses of female genital tract epithelium. Methods: Vaginal douche was collected from two groups, one group was healthy as control group, and the other had chlamydia trachomatis infection and then compared IL-8, TNF-α, HD-5 levels with ELISA method. With LPS to stimulate the normal endocervical epithelial ceils model,the supernatants were collected at 0,24,48hour,and used ELISA to see if there were any changes of TNF-α,HD-5 level. Results:IL-8,TNF-α,HD-5 levels from CT group were much higher than those from the control group,and the differences were statistically significant: IL-8 ( 136. 226 ±33. 561 vs 92. 294 ± 29.927)pg/mg,TNF-α(83. 344 ±21. 601 vs 59. 992 ±20. 106) pg/mg,HD-5 (0.531 ±0.261 vs 0. 368 ±0. 145 ) μg/ml. Meanwhile, in vitro experiment showed that the TNF-α, HD-5 levels increased after LPS stimulation in a time dependent manner. Conclusions: In vivo experiment shows that chlamydia trachomatis can change female genital tract local immunological conditions while in vitro experiment demonstrated that the normal cervical epithelium may take part into the process and play a critical role in such responses.
