小鼠乳腺中瘦素及其受体的表达与作用 被引量:5

摘要 瘦素(Leptin)是影响乳腺发育和泌乳的自分泌和旁分泌因子.为探讨瘦素在乳腺发育、泌乳和退化过程中的调控及作用机理,本研究应用激光共聚焦显微技术对小鼠乳腺发育、泌乳以及退化过程中瘦素及其长型受体(OB-Rb)蛋白水平的表达定位进行检测,并采用乳腺组织体外培养和Westernblot方法研究了妊娠期、泌乳期和退化期瘦素对乳腺的作用以及乳腺发育各个时期瘦素的信号转导通路.结果显示,小鼠乳腺整个发育过程中瘦素和OB-Rb的表达呈显著正相关,在青春期最高,妊娠期下降,泌乳期维持在低水平,幼仔离乳后乳腺退化,瘦素和OB-Rb的表达显著上调,并在乳腺退化13天恢复到青春期水平;瘦素及其受体的表达部位的变化显示瘦素专一性地诱导OB-Rb的表达,表明瘦素通过与OB-Rb共同作用调控乳腺发育和泌乳的生理机能;妊娠期,瘦素与OB-Rb结合,通过激活JAK-MAPK信号通路促进乳腺导管上皮细胞的增殖和分化;泌乳期,瘦素激活JAK-STAT5信号通路诱导乳腺中β-酪蛋白基因表达;退化期,瘦素激活JAK-STAT3信号通路启动乳腺细胞凋亡并参与乳腺组织重建过程.
