氯胺酮对白细胞表面L-选择素、β_2整合素的影响

摘要 目的研究氯胺酮对化学趋化因子(FMLP)刺激下白细胞表面L-选择素和β2整合素的影响.方法将人白细胞随机分为空白对照组(仅加(DMEM)培养液)、实验对照组(仅加FMLP)、K1-4组(氯胺酮的浓度分别为0.3μg/mL、3.0μg/mL、30μg/mL、300μg/mL),加入不同浓度的氯胺酮温育30 min后,加入FMLP 1μg/mL,作用20 min,用流式细胞仪测定白细胞表面L-选择素和β2整合素的变化.结果白细胞在FMLP的作用下,其表面β2整合素的表达和L-选择素的蜕落明显增加,≥30μg/mL的氯胺酮能抑制FMLP引起的β2整合素的表达上调和L-选择素的蜕落.结论氯胺酮可抑制FMLP刺激引起的白细胞的活化. Objective To tegrin of neutrophils by formyl investigate the effects of ketamine on the expression of L - selectin and β2 -- inmethi - onyl- leucy - phenylalanine (FMLP). Methods The neutrophils were randomly divided into blank control group (only DMEM culture fluid), experimental control group (only added FMLP) and K1 -4 group. Neutrophils were incubated with FMLP 1.0μmol/L for 20 minutes after the absence or presence of ketamine (0.3 μg/mL, 3.0 μg/mL, 30 μg/mL and 300 μg/mL) for 30 minutes. Subsequently, the expressions of L - selectin and β2 - integrin were obtained by flow cytometric analysis. Results The expression of β2 - integrin and shedding of L - selectin of neutrophils significantly increased by FMLP. Ket- amine (/〉30 tLg/mL) repressed the FMLP - induced upregulation of β2- integrin and shedding of L - selectin. Conclusion Ketamine may restrain the activation of FMLP - induced neutrophils.
