对产油脂酵母的细胞破碎方法及油脂提取效果的比较 被引量:28

摘要 研究了冻融破碎法、超声波破碎法、酸热法以及冻融和酸热结合法破碎发酵性丝孢酵母(Trichosporon fermentans)细胞,并考察了乙醚-石油醚法和氯仿-甲醇法萃取细胞内油脂.实验结果表明,采用酸热法破碎细胞后以乙醚-石油醚作为萃取溶剂油脂提取效果较好,油脂得率高达29.8%. Trichosporon fermentans were disrupted by freeze-meh, uhrasonic wave, acid-heating method and freeze-meh & acid-heating method respectively. The lipid extraction rates of Trichosporon ferments by the ether-benzine and chloroform-methanol were compared. The results implied that the ceils were disrupted by acid-heating and then extracted by the ether-benzine, by this procedure, the lipid extraction rate could reach higher level as high as 29.8%.
