几种贝类消化酶同工酶比较分析和遗传关系探讨

摘要 以缢蛏(Sinonovacula constricta)、剖刀蛏(Cultellus scalpellum)、文蛤(Meretrixlinnaeus)、栉孔扇贝(Chlamys farreri)、杂色鲍鱼(Haliotis diversicolor)等为研究对象,通过其蛋白酶、酯酶和淀粉酶的同工酶分析,表明3种消化酶在不同贝类中表达图谱各异、强弱有别。其中蛋白酶图谱最为简单,出现2条同工酶带;酯酶酶谱较为复杂,共出现8种同工酶带;淀粉酶酶谱最为复杂,总共出现9条同工酶带。相似度指数统计显示缢蛏与文蛤、剖刀蛏和栉孔扇贝之较高,而与杂色鲍鱼差异较大。进一步根据同工酶酶谱差异指数聚类,结果缢蛏和剖刀蛏首先聚在一起,然后文蛤、栉孔扇贝和杂色鲍鱼依次聚类,这一结果与参试5种贝类的生物学分类地位非常一致。 The genetic relations and the digestive isozymes in five shellfishes were compared and analyzed in this paper. The results showed that the different isozymes of proteins, esterase and amylase in Sinonovacula constricta ,Cultellus scalpellum,Meretrix linnaeus, Chlamys farreri and Haliotis diversicolor. Also the ranges of the quantifications were expressed. The smallest genetic variations of proteins in five shellfishes were given, and the amylases of visible genetic variations were done. According to the zymogram differentia index, the closest relation was found between Sinonovacula constricta and Cultellus scalpellum, Haliotis diversicolor was different from Sinonovacula constricta,Cultellus scalpellum,Meretrix linnaeus and Chlamys farreri.
