摘要
为了研究金银花质量控制的有效手段。通过建立反相高效液相色谱法测定了金银花中绿原酸的含量。测定条件为:色谱柱为Nucleosil C18柱(4.6×250m,5μm),流动相为甲醇-0.4%磷酸(体积比为25:75),流速1.0ml/min,光电二极管阵列检测器,检测波长327am,柱温25oC。绿原酸在0.64-6.40μg范围内其进样量与峰面积呈良好的线性关系(r=0.9996),平均回收率为99.3%,相对标准偏差(RSD)为0.8%。以6种不同浓度(0%-95%)的乙醇溶液为提取剂,对绿原酸的提取率进行考察的结果表明,30%乙醇溶液的绿原酸提取率最高,且杂质峰的数量明显减小。该方法具有简便、快速,回收率、精密度高的优点,可作为金银花质量控制的有效手段。
The high performance liquid chromatographic method was developed for the determination of chlorogenic acid in Flos Lonicerae. The chromato-graphic separation was carried out in a Nucleosil C18 column (4.6×250 mm, 5μm) with a mobile phase consisting of methanol-0.4 % H3PO4 (25:75, V/V) mixture.The detection wavelength of photodiode array detector was set at 327 nm and the column temperature was set at 25℃.The calibration curve was linear in the range of 0.64 - 6.40 μg for chlorogenic acid (r = 0.9996). the average recovery of chlorogenic acid was 99.3 % with a relative standard deviation (RSD)of 0.8 % .the method was simple,reliable and suitable for quality evaluation and control of Flos Lonicerae.
