摘要
目的 建立一种测定大鼠胃肠道内容物及肝组织中5-氟尿嘧啶浓度的方法。方法 取大鼠胃肠道内容物及肝组织匀浆液,加入内标利巴韦林后,用乙酸乙酯提取,60℃水浴挥干,流动相溶解进样。流动相为水-甲醇(体积比100∶1),色谱柱为ODS C18(4.6 mm×200 mm,粒径5μm),流速为0.8 mL·min^-1,检测波长为266 nm,柱温为室温。结果 方法回收率为90.2%~100.4%,提取回收率为58.5%~80.4%,日内RSD为4.2%~7.2%,日间RSD为4.7%~7.9%。结论 HPLC法可以用于大鼠胃肠道内容物和肝组织中5-氟尿嘧啶浓度的检测。
Objective To establish a method for the determination of 5-fluorouracil concentration in gastrointestinal contents and liver of mice. Methods Gastrointestinal contents and liver sample was extracted with ethyl acetate and organic phase was evaporated to dryness at 60℃. The residues were redissolved in mobile phase. The 5-fluorouracil was analyzed on ODS ClS column (4.6 mm × 200 mm, 5 μm) ; the flow rate was 0.8mL·min^-1;the detection wavelength was set at 266 nm; column temperature was room temperature. Results The method recovery was 90.2 % - 100.4 %, extract recovery was 58.47 % - 80.45 %. With-in precision (RSD) was 4.18% - 7.15%, between-day precision (RSD)was 4.72% - 7.87%. Conclusions The method was sensitive, reproductive for the determination of 5-fluorouracil concentration in gastrointesti- nal contents and liver sample.
