摘要
探讨了白鲫(Carassius auratus cuvieri)C反应蛋白(CRP)亲和层析法提取过程中,不同的硫酸铵饱和浓度、缓冲液pH值、缓冲液离子强度和配基浓度下对白鲫CRP的提取效率的影响.结果发现,在硫酸铵饱和浓度80%,缓冲液pH值8.5,缓冲液离子强度为0.05 mol/LTris以及配基浓度为8μmol/mL介质的条件下,白鲫CRP的提取效率最高.
This study probes into the optimization for CRP separation condition. CRP was extracted by affinity chromatography from crucian(Carassius a uratus cuvieri). The effects of ammonium sulfate degree of saturation, the buffer p H, ion intensity, the genin density on the yield of CRP were investigated. Results showed that the crudian CRP extraction efficiency was best when the condition was in 80% ammonium sulfate density,the buffer solution pH 8. 5,0.05mol/L Tris density in buffer solution as well as the 8 μmol/L genin in 1 mL medium.
