槟榔碱对α平滑肌肌动蛋白在口腔黏膜成纤维细胞中表达的影响 被引量:3

摘要 目的了解肌成纤维细胞与口腔黏膜下纤维性变(OSF)的关系并探讨肌成纤维细胞分化的来源和机制。方法体外培养正常口腔黏膜成纤维细胞和OSF成纤维细胞,用免疫组织化学方法和RT-PCR方法检测α平滑肌肌动蛋白的表达,并观察槟榔碱对其表达的影响,同时用ELISA方法检测了槟榔碱对成纤维细胞合成TGFβ1的影响。结果OSF来源的成纤维细胞中α平滑肌肌动蛋白表达明显高于正常口腔黏膜成纤维细胞,槟榔碱对体外培养成纤维细胞合成α平滑肌肌动蛋白和TGFβ1水平无明显影响。结论肌成纤维细胞可能在OSF的发病中起作用;OSF中肌成纤维细胞的来源可能与槟榔成分对成纤维细胞的直接作用无关。 [Objective] To evaluate the relationship of myofibroblasts and oral submucous fibrosis and to explore the origination and mechanism of the differentiation of myofibroblasts. [Methods] The fibroblasts from oral submucous fibrosis patients and normal controls were isolated and cultured. The expression of α smooth muscle actin was evaluated by immunoceUchemistry and RT-PCR, and the level of TGFβ1 was detected by ELISA. [Results] The expression of α smooth muscle actin in fibroblasts from oral submucous fibrosis was higher than that in normal control.No significant effect was observed in the α smooth muscle actin expression and the level of TGFβ1 between the fibroblasts stimulated by arecoline and the control. [Conclusion] Myofibroblasts may play a role in the pathogenesis of oral submucous fibrosis. The origination of myofibroblasts in oral submucous fibrosis may not be led directly by the stimulation of arecoline.
