清肝浸膏对实验性肝损伤模型细胞因子水平的影响

Effect of Qinggan Extract on the levels of cytokines of experimental liver injury models

目的研究清肝浸膏对实验性肝损伤模型细胞因子水平的影响。方法采用D-氨基半乳糖(D-GalN)诱导大鼠的急性肝损伤模型,用卡介苗联合脂多糖(BCG+LPS)诱导小鼠的免疫性肝损伤模型,观察清肝浸膏不同剂量组对血清谷丙转氨酶(ALT)、谷草转氨酶(AST)及细胞因子肿瘤坏死因子-α(TNF-α)、白介素-1β(IL-1β)、白介素-8(IL-8)和白介素-18(IL-18)活性的影响。结果清肝浸膏能明显降低肝损伤模型的转氨酶水平,并能降低D-氨基半乳糖所致急性肝损伤模型大鼠血清中的TNF-α、IL-1β、IL-8含量(P<0.01),也能明显降低BCG+LPS诱导的免疫性肝损伤模型小鼠血清中的TNF-α、IL-18含量(P<0.01)。结论清肝浸膏对实验性肝损伤模型有一定的保护作用,其保护作用的机制可能和降低细胞因子的水平有关。

Objective To study the effect of Qinggan Extract on the levels of cytokines of experimental liver injury models. Method The acute liver injury model was established by using D-galactosamine. The immunological liver injury model was established by using BCG and lipopolysaccharide （LPS）. Serum alanine aminotransferase （ALT）, aspartate aminotransferase （AST） and cytokines were determined. Results Qinggan Extract could decrease ALT and AST level in both of the liver injury models, and could also decrease TNF-α, IL-1 β and IL-8 levels in rats with D-galactosamine induced liver injury and TNF-α and IL-18 levels in mice with immunological liver injury. Conclusion Qinggan Extract has certain protective effect on experimental liver injury models. The mechanism perhaps is related to the reduction of cytokines.
