摘要
[目的]为了获得高纯度鸡免疫球蛋白(Ig),并保持其生物学活性。[方法]比较几种纯化鸡血清免疫球蛋白(IgG)的方法。用饱和硫酸铵(NH4)2SO4沉淀法或乙醇沉淀法提取鸡血清IgG,经透析除盐,将初提物过sephadexG-200柱进一步分离IgG,经免疫电泳和SDS-PAGE电泳进行纯度鉴定。[结果]结果表明:在该试验条件下,(NH4)2SO4盐析和乙醇沉淀法获得的鸡IgG经sephadexG200柱分离获得高纯度的鸡IgG。[结论]该研究为找到较好的纯化鸡血清IgG方法提供了试验依据。
The objective of the study is to obtain highly purified chicken IG and maintain its biological activity.Some methods about purification of serum IgG from chicken were appraised.Firstly,chicken serum was extracted by saturated ammonium sulfate precipitation or ethanol precipitation.After dialysis for desalination,the original extracts are purified by sephadex G-200 column chromatography.The purified chicken IgG was identified by SDS-PAGE.The study has found a fairly good method in obtaining purified chicken serum IgG and provided the basis for further experiment.
