Detection of early apoptotic spermatozoa in normal human semen

目的通过细胞化学方法观察正常精液中精子早期凋亡发生的程度。方法采用荧光标记的AnnexinV和碘化丙啶对34例具有正常精液分析参数的精子进行染色和显微观察,计数凋亡精子发生率。结果精液中早期凋亡精子发生率为6.5(4.9,7.4)%,个体间存在较大的差异。凋亡精子发生率和年龄和禁欲时间等不相关。结论精液中存在少量的早期凋亡精子且个体间差异较大,荧光标记的AnnexinV和碘化丙啶染色显微镜观察可以有效地对其进行量化观察。

Objective To observe the sperm early apoptosis in the normal human semen by using cytocbemical staining method. Methods Sperm from 34 semen with normal parmneters of semen analysis was microscopically observed by fluorescein-labeled AnnexinV and Proidium iodid staining,and the percentage of apoptoctic sperm in the samples was counted.Results Apoptotic sperm occurred in the samples was 6.5 （4.9,7.4） %, and there was an interindividual difference obviously. Apoptotic sperm rate was not significantly correlative with age and abstinence days. Conclusion There is a low population of early apoptotic sperm in human ejaculate, and an obvious individual difference of the apoptesis occurrence was found in this study. The fluorescein-labeled AnnexinV and PI microscopic observation could be efficiently used for the sperm apoptosis quantitive analysis.
