利福霉素钠与异烟肼合用对小鼠肝脏毒性的实验研究 被引量:4

Empirical Study on the Hepatotoxicity Induced by Rifamycin Sodium Plus Isoniazid in Mice

摘要 目的:比较利福霉素钠与异烟肼单用及合用的肝脏毒性。方法:将40只小鼠完全随机分为4组,即生理盐水组、利福霉素钠组、异烟肼组、利福霉素钠与异烟肼联合用药组,分别腹腔注射和灌胃给药8d后摘眼球取血处死小鼠,测定血清丙氨酸氨基转移酶(ALT)和天冬氨酸氨基转移酶(AST)的活性,计算肝指数,用光镜观察肝脏组织细胞病理学的改变情况。结果:各药物组的肝指数和血清ALT、AST活性水平均较生理盐水组高(P<0.0001~P<0.05);病理组织学观察显示2药均能引起肝细胞的变性和坏死,尤其是合用组更明显。结论:利福霉素钠和异烟肼对肝脏均有损害作用,且合用后肝脏毒性加重。 OBJECTIVE: To compare the hepatotoxicity which induced by rifamycin sodium and isoniazid by alone or combination in mice. METHODS: Forty mice were divided randomly into 4 groups: the sodium chloride group, the rifamycin sodium group, the isoniazid group and the drug - combination group. The drugs were administered to mice by i.p.or i.g .once daily for 8 days. Each mouse was killed by ophthalmectomy. The blood was collected and the liver was excised immediately .The activity of serum ALT and AST was measured and the liver index in mice was calculated. Light microscope was used to observe the histopathological changes of the hepatic cells. RESULTS: The liver index and the activity of serum ALT and AST increased in all groups expect the sodium chloride group(P〈0.000 1-P〈0.05) . Histopathological examination showed that rifamycin sodium and isoniazid could cause the degeneration and necrosis of hepatic cells, especially in the drug - combination group. CONCLUSION: Rifamycin sodium and isoniazid could induce liver damage in mice. The degree of liver damage were more serious in the drugcombination group than those in the single drug group.
