妊娠高血压综合征患者血清肿瘤坏死因子-α水平及胎盘组织缺氧诱导因子的表达

The Level of Secrum TNF-αand the Expression of Hypoxia Inductive Factor of Placenta Tissue in Patients with Pregnancy-induced Hypertension Syndrome

目的了解妊娠高血压综合征(妊高征)患者血清肿瘤坏死因子(TNF-α)的水平变化及胎盘组织缺氧诱导因子-1α(HIF-1α)的表达情况,探讨二者在妊高征发病中的作用及相互关系。方法采用放射免疫法测定107例妊高征患者(轻度33例、中度34例、重度40例)及100例正常孕妇的血清TNF-α水平。胎盘娩出后取胎盘组织经过固定、切片,采用免疫组化法检测妊高征患者和正常孕妇胎盘组织中缺氧诱导因子-1α的阳性表达率。结果妊高征患者和正常孕妇血清TNF-α水平间差别有显著性意义(P<0.01),轻度妊高征患者与正常孕妇血清TNF-α水平间差别无显著性意义(P>0.05)。妊高征患者与正常孕妇胎盘组织缺氧诱导因子-1α的阳性表达率间差别有显著性意义(P<0.05),且其阳性表达率随妊高征患者的病情加重而升高。结论(1)妊高征患者血清TNF-α水平及HIF-1α明显高于正常孕妇,且随妊高征患者病情的加重,TNF-α水平和HIF-1α阳性表达率明显升高,说明二者在妊高征发病中起重要作用。(2)血清TNF-α与胎盘组织缺氧诱导因子-1α呈正相关。

Objective To contribute to the knowledge of changes in the level of secrum TNF - α and the expression of hypoxia inductive factor （ HIF - 1 α） in patients with pregnancy - induced hypertension syndrome （ PIH）, and to find out their interrelationship in PIH. Methods The levels of secrum TNF - α in 107 patients with PIH （33 mild, 34 moderate, 40 severe） and that of 100 normal pregnant women were measured by radioimmunoassay. The positive expressions of hypoxia inductive factor （ HIF - 1α） in both normal pregnant women and patients with PIH were measured in an immuno - histochemical method. Results The patients with PIH was significantly different from the normal pregnant women in secrum TNF-α （ P 〈 0. 01 ）, no significant difference was found in secrum TNF - α between the mild cases and the normal pregnant women （ P 〈 0. 05 ）, there was significant difference in positive expression of hypoxia inductive factor （HIF- 1α） of placenta tissue between the patients with PIH and the normal pregnant women, and the positive expression in patients with PIH was increased when their condition became more serious. Conclusion The levels of secrum TNF - α and HIF - 1 α in patients with PIH are significantly higher than that of normal pregnant women, and there will be a significant increase in positive expression of TNF - α and HIF - 1α when the patients＇ condition becomes serious. The results demonstrate that both of them play an important role in PIH, and secrum TNF - 1α has positive correlation with hypoxia inductive factor of placenta tissue
