肺癌组织中WISP-1 mRNA和蛋白的表达

摘要 目的:探讨人肺癌组织中WNT诱导的分泌型蛋白-1(WISP-1)基因的表达与临床病理因素的关系。方法:采用real-timePCR和免疫组化法检测原发性肺癌患者的癌组织和癌旁正常肺组织中WISP-1mRNA和蛋白的表达。结果:肺癌组织和正常肺组织中WISP-1mRNA的相对表达量分别为(-3.09±3.53)μg/L和(-5.82±3.67)μg/L,WISP-1蛋白阳性率分别为63.3%和39.2%,肺癌组织中WISP-1mRNA的相对表达量和蛋白阳性率均高于正常肺组织(P均<0.05)。肺癌组织中WISP-1mRNA的表达与病理学类型、年龄有关(P<0.05)。多元线性回归分析显示:病理类型、年龄、家族史、转移、性别等对WISP-1mRNA的表达有影响。结论:WISP-1基因可能是原发性肺癌发生发展过程中的重要生物学标志物,WISP-1基因表达的蛋白产物可作为临床干预治疗的重要靶蛋白。 Aim : To investigate WISP-1 gene expression level and its relation to clinical and pathological factors in human lung tumors tissue. Methods :The mRNA and protein expression levels of WISP-1 in samples from primary lung tumor and their matched normal lung tissue were determined using real-time PCR assay and immunohistochemistry staining, respectively. Results: The mRNA expression levels of WISP-1 in lung cancer tissue and the normal lung tissue were ( - 3.09 ± 3.53 ) μg/L and ( - 5.82 ± 3.67 ) μg/L, and the positive expression rates of WISP-1 protein were 63.3% and 39.2%. The mRNA expression levels and positive rates of WISP-1 protein in lung cancer tissue were both higher than those in normal lung tissue (P 〈 0.05). There was association between WISP-1 mRNA expression of WISP-1 and tumor pathology types and age of patients (P 〈 0.05). Multiple linear regression analysis revealed that tumor type, age, and family history, lymph node metastasis and sex were valuable predictors for WISP-1 mRNA expression. Conclusion: It is suggested that WISP-1 may play an important role in the progression of primary lung cancer and the proteins might serve as valuable targets for therapeutic intervention.
