摘要
目的:探求光敏剂5-氨基酮戊酸(ALA)口服后在膀胱中的代谢,寻求光敏诊断最佳的ALA口服剂量及时间。方法:予昆明小鼠口服不同剂量的ALA(20mg/kg、40mg/kg、60mg/kg8、0mg/kg 4个组别),不同时间(1小时、2小时、4小时、6小时、8小时、10小时、12小时、24小时、48小时)以激光诱导荧光OMA检测系统测定膀胱卟啉和组织荧光强度,并经数据处理观察ALA不同时间在膀胱中的代谢状态。结果:膀胱卟啉荧光峰值除以组织荧光峰值的比值在口服ALA40 mg/kg后1小时最大。结论:从实验数据分析口服ALA作OMA荧光光谱分析以40mg/kg为最佳检测剂量,最佳检测时间为口服ALA后1小时。
Objective: To research on the metabolism of ALA taking orally in bladder of normal mice and seek suitable dose and diagnostic time of ALA taking orally. Methods: Different dose of ALA taking orally(20mg/kg,40mg/kg ,60mg/kg,80mg/kg), different time after ALA taking orally(lh,2h,4h,6h,8h,10h,12h,24h,48h), the fluorescence of bladder and tissue was measured when irradiated by laser. By analyzing the spectrum of fluorescence with optical multi-channel analyzer (OMA) the suitable dose and diagnostic time for tumor detection were obtained. Result:The ratio of Ⅰbladder/Ⅰtissue is largest for 40mg/kg and l h after ALA taking orally. Conclusion:Measuring fluorescence spectrum of bladder and tissue and analyzing the spectrum of fluorescence with optical multi-channel analyzer (OMA), the most suitable dose was 40mg/kg of ALA taking orally, and the measure time is taken 1 hour after ALA taking orally
