大肠杆菌hemA基因的高效表达及对5-氨基乙酰丙酸合成的影响 被引量:1

摘要 大肠杆菌(Escherichia coli)中的hemA基因编码谷氨酰tRNA还原酶,该酶是E.coli生物合成5-氨基乙酰丙酸(5-ALA)途径中的限速酶。本试验在pEXT20-hem AE.coliBL21(DE3)表达载体的基础上,从质粒克隆hemA基因克隆到载体,构建表达载体pET28a-hemA,以探讨更佳的重组大肠杆菌hemA基因的表达效果。对其蛋白质进行纯化,测得发酵菌液上清液中5-ALA含量达到42.5 mg/L;并对上清提取液中卟啉类物质的积累量进行初步测定,发现403 nm处吸收峰值明显高于同条件发酵提取pEXT20-hemAE.coliBL21(DE3)重组菌。试验表明,用表达载体可以更好地表达hemA基因。
