摘要
目的探讨α-内收蛋白基因变异(Gly460Trp)与大动脉弹性相关性研究。方法(1)采用Complior SP脉搏波速度测定仪测量股-颈动脉PWV(C-FPWV),共319例,其中C-FPWV异常组204例,对照组115例;(2)采用聚合酶链反应和限制性酶切的片段长度多态分析方法检测α-内收蛋白基因变异(Gly460Trp)。结果(1)C-FPWV升高组收缩压(SBP)、舒张压(DBP)均显著高于对照组(P<0.05);(2)C-FPWV升高组WW、W等位基因频率显著高于对照组(P<0.05);(3) WW基因型与WG+GG基因型比较C-FPWV、收缩压、舒张压差异有显著性(P<0.05);(4)Logistic回归结果显示:α-内收蛋白460WW基因型与大动脉弹性有密切相关性(P=0.001,OR=3.234,95%CI1.655- 6.320)。结论α-内收蛋白基因变异(Gly460Trp)与大动脉弹性有密切相关性,WW型是大动脉弹性减退的敏感基因型。
Objective To assess the association beween -Adducin Gene polymorphism and big artel7 elasticity. Methods ( 1 ) Big ariel7 elasticity was measured with Complior SP automatic pulse wave measurement systenl of pulse wave velocity(PWV) in 319 sui)jeets,including 204 subjects with an abnormal C-FPWV and 115 normal controls. (2)The genetypes were determined with polymerase chain reaction and allele-specific reaetion enzyme analysis. Result ( 1 ) SBP and DBP in abnormal CFPWV were significantly higher than in normal CFPWV ( P 〈 0.05 ). (2) The frequencies of W allele and WW genotypes in abnormal C-FPWV group were significantly higher than in control group (P 〈 0.05 ). (3)C-FPWV ,SBP and DBP in WW genotype were significantly different from that in WG + GG ( P 〈 0.05 ). (4) Logistic regression analysis revealed that WW genotypes were associated with abnormal C-FPWV ( (P = 0. 001, OR = 3. 234,95% CI 1. 65545. 320). Conclusion The n α-Adducin Gene pnlymorphism is associated with big artery elasticity. WW genolype is a sensitive genolype reflecting the abnormality of big arterial compliance.
