鱼腥草素同系物对巨噬细胞膜蛋白荧光特性的影响研究(英文)

摘要 研究了鱼腥草素同系物(HOU-Cn)对巨噬细胞膜蛋白构象的影响.荧光分析表明鱼腥草素在浓度低于1.6×10-4mol/L范围内,随着浓度的上升,能迅速地淬灭酪氨酸残基的发射荧光强度.每个巨噬细胞可以结合1.70×1017分子的HOU-Cn,且随着碳链的延长,结合数略有增加.结果表明HOU-Cn通过疏水作用与巨噬细胞膜蛋白结合,由此改变膜蛋白的构象和激活巨噬细胞. The effects of houttuyfonate homologues (HOU-Cn) on the protein conformation of macrophage were investiga ted. The fluorescence experiments indicated that within the concentration range of 1.6 × 10^-4 mol/L (50μg/mL) or lower, HOU-Cn could quench the fluorescence of Tyr (tyrosine) residues quickly. Each macrophage could bind 1.70×10^17 molecules of HOU-Cn, and the binding number increased slightly with the carbon chain of HOU-Cn. It suggested that the combi nation between HOU-Cn and membrane proteins of macrophage was caused by hydrophobic interaction, which changed the conformation of membrane proteins and acffvated macrophage consequently.
