摘要
目的:建立了雷公藤多苷中雷公藤内酯甲的含量测定方法。方法:雷公藤多苷经柱层析洗脱后,采用HPLC法进行测定。色谱柱:C18柱,流动相:乙腈-水(75∶25),流速:1.0m1.min-1;检测波长为210nm。结果:雷公藤内酯甲与其他成分分离良好。线性范围为6～120μg.ml-1(r=0.9997)。平均回收率为98.98%,RSD为1.99%(n=9)。结论:方法简便、准确、重复性好,可用于雷公藤多苷的质量控制。
Objective ：To develop a RP-HPLC method for the determination of Wilforlide in Tripterygium glycosides. Methods ： The rough-preparation was eluted with ethyl acetate-acetone （3 ： 1） on a neutral alumina cartridge. Then, samples were achieved and separated by C18 column （250mm）〈 4.6mm, 5μm）with acetonitrile-water （75 ： 25） as mobile phase. The flow rate was 1. 0ml · min^-1. The UV detector was set at 210nm. Result ： Wilforlide was well-separated. The linear calibration curve was obtained in the concentration range of 6- 120μg ·mL^-1 （r = 0. 9997）. The average recovery of Wilforlide was 98.98%,RSD 1.99 % （n = 9）. Conclusion：The method is simple,accurate and reproducible. It is also proposed to be used for the quality control of Tripterygium glycosides.
