碳纳米管对小鼠肝和肺细胞DNA损伤的彗星实验研究 被引量:7

摘要 目的检测碳纳米管进入小鼠体内后对小鼠肝细胞和肺细胞DNA的损伤作用。方法将小鼠分别用不同剂量多壁碳纳米管(0.1、0.2、0.4mg/ml)暴露14天,每次0.2ml,每隔24h暴露1次,用单细胞凝胶电泳技术检测肝细胞和肺细胞DNA的损伤程度。结果0.1mg/ml剂量大小的碳纳米管可造成小鼠肝细胞和肺细胞DNA的严重断裂(P<0.01),而0.2、0.4mg/ml剂量则引起了明显的交联作用。结论碳纳米管可造成机体内肝细胞和肺细胞DNA的断裂和交联。 Objective To explore the hepatic and pulmonary cellular DNA damage after the mice injected with carbon nanotubes. Methods Mice were respectively exposed to different dosages of multi -walled carbon nanotubes (0.1,0.2,0.4mg/ml)for 14d, 0.2ml for every time and once for 24h. SCGE (single cell gel electrophoresis, comet assay) was performed to detect DNA damages. Results The hepatic and pulmonary cellular DNA strands breaks at 0.1 mg/ml ( P 〈 0.01 ) while obvious crosslinks were induced at 0.2 and 0.4 mg/ml. Conclusions Carbon nanotubes can induce DNA strands breaks and crosslinks of hepatocytes and pulmonary cells in vivo.
