摘要 目的探讨建立幼年大鼠缺氧缺血性脑病(HIE)动物模型的可靠方法。方法结扎幼年wistar大鼠(1月龄左右)左颈总动脉两端,置于8%浓度的低氧环境中2.5 h,观察大鼠行为;经心脏灌流4%多聚甲醛固定取脑,观察脑部外观变化,采用HE染色及光镜技术观察其脑组织结构改变。结果造模后幼鼠行为表现符合经典缺氧缺血性脑病动物模型的行为改变;缺氧缺血后3 h左侧大脑出现轻度脑损伤,24 h病变明显,4 d出现胶质细胞增生,18 d神经元大量坏死、丢失,多部位形成胶质瘢痕。病理变化与幼儿HIE相似。结论本方法建立的大鼠缺氧缺血性脑病动物模型快速、可靠,可用于实验研究。 Objective It is to study the reliable method for establishing young rat model of hypoxic-ischemic encephalopathy (HIE). Method The ends of left common carotid artery vascular of young Wistar rats (about one month old) were ligated, and put in 8 % of the hypoxic environment for 2.5 hours. Changes of behavior were observed. The appearance and pathological changes of rat brain were observed by light microscopy techniques, after 4 % paraformaldehyde was perfused in cardiac to fix brain, and HE staining was used. Results The behavior of young Wistar rat after operation was consistent with the behavior change of classic animal model of hypoxic-ischemic encephalopathy, Left cerebral hypoxic-ischemic brain was injuried slightly after 3 hours, more obvious after 24 hours, there were gliosis after 4 days, and there was a large number of neuron necrosis and loss and multisite glial scar after 18 days. The pathological changes was similar to the HIE of young children. Conclusion The method to make hypoxic-ischemic encephalopathy animal model is fast and reliable, so it can be used in experimental study.
