摘要
目的观察芪丹通脉片预处理对缺血再灌注大鼠心肌细胞损伤的影响。方法雄性SD大鼠随机分为4组,即假手术组(A组)、缺血再灌注模型组(B组)、芪丹通脉片加缺血再灌注Ⅰ组(C组)、芪丹通脉片加缺血再灌注Ⅱ组(D组)。大鼠麻醉开胸,结扎冠状动脉前降支40min,再灌注4h。采用Evan’sBlue和TTC染色测定心肌梗死范围,并TUNEL法测定大鼠心肌组织的细胞凋亡,检测血清中肌酸激酶(CK)和肌钙蛋白I(cTnI)的水平。结果不同剂量(1.08g/kg,3.24g/kg)芪丹通脉片预处理组,缺血/再灌注后心肌梗死面积和凋亡指数显著小于模型组(P<0.05或P<0.01);与假手术组比较,缺血/再灌注4h后B组大鼠血清CK和cTnI有统计学意义(P<0.05或P<0.01);而不同剂量芪丹通脉片预处理组再灌注后血清CK和cTnI含量明显降低,与B组比较有统计学意义(P<0.05或P<0.01)。结论芪丹通脉片预处理能够有效地抑制缺血/再灌注所致大鼠心肌损伤。
Objective To investigate the effect of Qidantongmai tablet (QDTMT) on myocardial ischernia injury in rats. Methods Male Sprague- Dawley rats were dMded randomly into 4 groups: Sham group, ischelnia/reperfusion (control group), QDTMT- I (1.08 g/kg) treatment group and QDTMT- Ⅱ (3.24 g/kg) treatment group. The rats were anesthetized and then followed by 40 min of myocardial ischemia and 4 h reperfusion. The protective effect of QDTMT was asscssed by tile infact size by Evans Blue and lriphenyltctrazolium chloride staining. The cardiomyocyte apopto,is was determined by TUNEL method and the level of creatine kinase(CK) and cardiac troponin Ⅰ (cTn Ⅰ) in serum. Results Following pretreatment with QDTMT at different doses, the ratio of infarct size, area at risk and cardiomyocyte apoptosis indexes reduced and they were significantly smaller than that in control (P〈0.05 or P〈0.01 ). The levels of CK and cTnI in control were significantly higher than that in sham group. The levds of CK in groups QDTMT trcatmcnt at doses1. 08 g/kg or 3.24 g/kg were significantly lower than that in control ( P 〈 0.05 or P 〈 0.01). Conclusion Pretreatment with QDTMT could protect heart against ischemia reperfusion injury .
