摘要
目的研究ag85b-卡介苗重组疫苗的免疫原性。方法以生理盐水、卡介苗、ag85b-卡介苗重组疫苗免疫小鼠,采用ELISA法检测小鼠血清中特异性抗体,MTS法分析小鼠脾淋巴细胞增殖指数。结果ag85b-卡介苗重组疫苗组小鼠体重变化与对照组相比,无显著性差异。应用不同抗原检测各组小鼠不同时间采集的血清,ag85b-卡介苗重组疫苗组、卡介苗组均有抗Ag85B特异性抗体产生,ag85b-卡介苗重组疫苗组抗Ag85B特异性抗体水平高于卡介苗组;卡介苗、ag85b-卡介苗重组疫苗免疫后15d,抗体水平已有升高,45d达到最高水平。用不同抗原刺激实验组及对照组小鼠脾淋巴细胞,平均刺激指数均达2.0以上。结论结核分枝杆菌ag85b基因在卡介苗中能够表达特异的蛋白,刺激小鼠产生特异性抗体。
Objective To evaluate the immunity of ag85b-recombinant BCG vaccine. Methods The antigen-specific antibody levels of the mice immunized with ag85b-recombinant BCG were detected by ELISA, and multiplication of mouse lymph-cells was analyzed by MTS. Results There was no difference in weight between the mice immunized with saline and BCG, the control groups and those immunized with ag85b-recombinant BCG vaccine, the experimental group. After the serum of mice in each group collected at different time were detected with antigens such as conA, ESAT6, MPT64, Ag85A, Agg5B protein and PPD, anti-Ag85B specific antibody was found in the mice immunized with ag85b-recombinant BCG vaccine and in those immunized with BCG, with the antibody levels of the former group higher than those of the latter group. The antibody levels of BCG and the experimental group came to rise on the 15th day and reached the peak on the 45th day after immunization. When the mouse lymph-cells of the experimental group and the control groups were stimulated by different antigens, the mean stimulation index came to more than 2.0. Conclusion ag85b-recombinant BCG vaccine, antigen-specific gene of mycobacterium tuberculosis, can express specific proteins in BCG and stimulate B cell of mouse to produce specific antibody.
